When Ellen DeGeneres came out as gay 17 years ago, the unexpected professional backlash and personal isolation that followed was a rude awakening.
"I thought everyone knew me and I didn't think that one little adjective was going to define me," DeGeneres, 56, exclusively tells PEOPLE in the latest issue. "I didn't have perspective for a while."
But these days, with a hugely successful daily talk show, best-selling books and an upcoming gig as the host of the 86th Annual Academy Awards on March 2, that painful rough patch is a distant, if surreal, memory.
"Now I look at it as a movie that I saw that someone went through," she says, "and I only experience the amazing life that I have right now."
One big reason for DeGeneres' gratitude: her thriving marriage to actress Portia de Rossi, whom she wed in 2008. "I love her so much it kills me," says DeGeneres.

Ellen DeGeneres wants the world to know that despite reports to the contrary, she's "happily married" to Portia de Rossi.
The talk show host addressed rumors about their five-year marriage during a hilarious monologue on The Ellen DeGeneres Show on Wednesday, that began with a trip to the hair salon.
"The other day I'm at the hair salon getting these extensions," DeGeneres joked. "And there's tabloids all over the hair salon – that's where they are, I guess. All of a sudden I notice one sitting on the counter in front of me [and] it caught my eye because Portia and I were in the top right-hand corner and I thought, 'Oh, that's sweet, they're honoring us.' "

Ellen DeGeneres and wife Portia de Rossi are both vegan, but that doesn't stop them from enjoying a Thanksgiving feast.
"It doesn't make [Thanksgiving] harder at all. It makes it easier on the turkeys, too. They get to live," DeGeneres, 55, told PEOPLE at the Duracell Power a Smile and Toys for Tots event on Nov. 22.
So what does The Ellen DeGeneres Show host fill her plate with?
"Yams, green beans, stuffing ... any kind of vegetables. Stuff like that," she said.

Ellen DeGeneres is so happy she could dance.
"It's a supremely wonderful day for equality," the talk show host Tweeted on Wednesday. "Prop 8 is over, and so is DOMA. Congratulations everyone. And I mean everyone."
The Supreme Court earlier handed a major victory for gay rights in two decisions. It cleared the way for the resumption of same-sex marriage in California by letting stand earlier court declarations that Proposition 8 was unconstitutional, and struck down the Defense of Marriage Act provision denying federal benefits to married gay couples.

A little too friendly?
That was the scene for Jennifer Aniston when she sought some impromptu hosting advice from her former Friends costar Matthew Perry in a funny new skit to promote her Wednesday gig as co-host of The Ellen DeGeneres Show.
In the clip (watch it below), Aniston knocks Perry's door to get some tips from her former castmate, who also has co-hosted the show.
"We haven't actually seen each other in like a really long time, 8 years. ... You really should have called," said a nervous Perry, who looked on sheepishly as their mutual pal and former costar Courteney Cox emerges from his house after an obvious sleepover.
